Diff-Quik® Evaluation of Round Cells and Sperm

Abstract

Diff-Quik is a proprietary brand of a Romanowski-type stain that consists of a light blue fixative (triarylmethane dye), a red stain (eosin, xanthene-type dye), and a blue stain (thiazine dye). Diff-Quik stain has been used primarily to assess round cells in a semen sample, but may also be used, with some modification in staining technique, to assess spermatozoa. Staining with a Diff-Quik stain offers an alternative to the more common eosin-nigrosin stain for the evaluation of sperm morphology. This chapter discusses the step by step procedure to assess the round cells and spermatozoa with illustrations. It outlines the necessary conditions to be followed for the Diff-Quik evaluation of round cells and sperm.
