Diff-Quik is a proprietary brand of a Romanowski-type stain that consists of a light blue fixative (triarylmethane dye), a red stain (eosin, xanthene-type dye), and a blue stain (thiazine dye). Diff-Quik stain has been used primarily to assess round cells in a semen sample, but may also be used, with some modification in staining technique, to assess spermatozoa. Staining with a Diff-Quik stain offers an alternative to the more common eosin-nigrosin stain for the evaluation of sperm morphology.
