Calibrated Spectrophotometer Evaluation of Sperm Concentration

Abstract

Determination of spermatozoal concentration is required for the accurate calculation of a breeding dose and subsequent dilution of semen for cooled semen transport. Sperm concentration may be determined via a number of methods including manual counting with a hemocytometer, spectrophotometer, computer aided microscopy, and fluorescent assay techniques. Calibrated spectrophotometers dedicated to estimating stallion sperm concentration are commercially available. This chapter discusses the involved in evaluation of sperm concentration using a cuvette, microcuvette, and chip. Spectrophotometric estimation of sperm concentration can only be done using raw semen as opaque extenders used for cooled transport or cryopreservation will alter light transmission. A manual hemocytometer count can be performed in the event that a spectrophotometric determination of sperm concentration is in question.
