Several laboratories now perform cytotoxicity assays at several lymphocyte-to-target cell ratios by adding increasing numbers of lymphocytes to a fixed number of sl Cr-labeled target cells and express results graphically or in terms of lyric units, where one lytic unit is the number of lymphocytes required to effect a chosen percent cytotoxicity (Cerottini and Brunner, 1971; Kay et al., 1977). N2 - The construction and use of a simple semiautomatic device for collecting equal aliquots of supernates after radioisotope release assays of cellular cytotoxicity are described. This device simultaneously removes an aliquot of the supernate from each of the 12 wells in one row of a microtiter plate, and transfers these aliquots directly to disposable test tubes for insertion into a γ-counter. Results obtained using this device were more reproducible than manual methods for supernate collection.
